Description

Low Molecular Weight Protein Marker is a mixture of six purified proteins (14 to 94 kDa). This unstained protein marker can be detected by Coomassie Brilliant Blue R-250 or silver stains after run on SDS polyacrylamide gel.

Protocol

  1. Thaw the protein marker at room temperature. Do not boil it.
  2. Mix the tube gently and load an appropriate volume of the protein marker on to the gel.
    • Mini-gel: 5 μl per well (0.75−1.0 mm thick) or 10 μl per well (1.5mm thick)
    • Large-gel: 10 μl per well (0.75−1.0 mm thick) or 20 μl per well (1.5 mm thick)

Notes

  • It is not necessary to pre-heat the protein marker prior to loading on a gel.
  • It is recommended to use 12−15% SDS-PAGE gel for electrophoresis. Use high-percentage gels (15%) to obtain well-resolved bands on low-molecular weight proteins.
  • For silver stain applications, dilute the protein marker approximately 1/50 in reducing sample buffer.
  • If additional bands appear in the protein ladder add newly prepared dithiothreitol (DTT) solution to 100mM final concentration. DTT oxidation in the storage buffer can cause the appearance of additional bands.
  • Shelf Life 1 year at −20°C.
